My thumb is D.Y.I.N.G due to RT-PCR

Yes.... all the pipetting is really killing my thumb... the meat i mean... flesh... is feeling really sore.... this sucks... and i looked at the booking sheet for the realtime PCR machine carefully today... and to my horror.... my mentor booked a time slot for every day until the end of next week...

nooooooooooooooooooooooooooooooooooooooooooooooooooooooooo!!!!!!!!!!!

that means more pipetting.... more master mixes to prepare... more (mix + short spin) X 2 for all the primers... more primer dilution if any new ones arrive... and argh... NO TALKING when doing/ preparing any PCR stuff....

well... but still... think positive!!! at least it's not the other RT-PCR.... the other RT-PCR is reverse transcription polymerase chain reaction... there is no need to use a 96 well plate... just use the strip tubes (8 small pcr tubes stuck together in a strip form)... but the reagents to add are like... argh.. so many... and must also take them out of -20C freezer and thaw them... and keep on ice... etc.... and after that... just put in into the stupid RT-PCR machine and click the settings and can come back in about 2 hours after it's done.... but the downside is that we need to cast agarose gel to run the samples later.... argh... the horror of ETBR (ethidium bromide)!!!!

after the 2 hours.. just add loading dye... mix... and load the samples into the wells in the agarose gel... then run the gel for 40 mins... the come back and go to the UV lamp there to view/ take picture of the gel... yeah.. that's about it... it's very tedious... much more compared to realtime PCR... so yeah.. i prefer resltime pcr...
